Baculovirus Particles (BVPs) for Antibody Polyreactivity Screening

Polyreactivity, defined as non-specific binding of an antibody to structurally unrelated targets, is a well-documented liability in therapeutic antibody development. Elevated polyreactivity has been associated with faster in vivo clearance of therapeutic antibody candidates in preclinical studies, and is commonly assessed at the lead-selection stage as one input to a broader developability evaluation.

Curia supplies purified baculovirus particles (BVPs) as a reagent for use as the coating antigen in an ELISA-based polyreactivity assay. Test antibodies that produce elevated absorbance against immobilized BVPs, relative to defined controls, are flagged for further evaluation and can be triaged, engineered, or advanced with additional monitoring.

Background

The reagent was originally offered by LakePharma, which was acquired by Curia in 2021 and has remained the gold standard for antibody polyreactivity testing. It has been referenced in peer-reviewed studies covering monoclonal, bispecific and single-domain (VHH-Fc) antibody formats, including in a 2024 study that used BVP-ELISA data as a training input for protein language model-based polyreactivity prediction.
